摘 要:目的建立并优化从马兜铃药材中提取马兜铃酸和的静态加压溶剂提取方法,考察各因素对提取效率的影响,并测定马兜铃、关木通、青木香和广防己中两成分的量。方法以马兜铃酸和的量为指标,采取单因素考察的方法,分别优化提取溶剂、药材粒径、提取温度、提取压力、提取时间、冲洗体积、循环数目、药材加入量等影响因素。结果优化后的条件为:甲醇提取,药材粉末粒径100~120目,提取温度120℃,提取压力10.3MPa,静态提取时间10min,冲洗体积40%,1个循环,药材加入量1.00g。结论本法可一次将马兜铃酸和从马兜铃等4种药材中完全提取,显示了良好的适用性。经比较,该法比传统的超声和索氏提取方法提取效率更高,节约溶剂,省时省力;静态加压溶剂法更适合于青木香中马兜铃酸的提取。Objective To optimize static pressurized liquid extraction (PLE) method for the extraction of aristolochic acids Ⅰ and Ⅱ (AAⅠ and AA Ⅱ ) from Fructus Aristolochiae and study the influences of re- lated factors. Methods The univariate design was introduced. The operational parameters, such as the type of solvent, particle size of the sample powder, extraction temperature, pressure, static time, flush volume, the number of cycles, and the amount of sample were optimized. Results The optimized result employed methanol as extraction solvent, particle size of 100--120 meshes, extraction temperature of 120 ℃, extraction pressure of 10.3 MPa, static time of 10 min, flush volume of 40%, 1 cycle, and sample amount of 1.00 g. The method was applied for four species of traditional Chinese medicinal materials including Fructus Aristolochiae, Caulis Aristolochiae Manshuriensis, Radix Aristolochiae, and Radix Arsitolochiae Fangchi. Conclusion This method can be used to completely extract AA Ⅰ and AA Ⅱ from Fructus Aristolochiae in once extraction. The comparison shows that this static PLE method is better than ultrasonication and Soxhlet methods with higher extraction efficiency and less time-consuming. It is also better than the dynamic one in the extraction of AAs from Radix Aristolochiae.
