宁心红杞胶囊对围绝经大鼠卵巢局部自分泌及旁分泌因子的调节作用

摘　　要：目的探讨宁心红杞胶囊对围绝经大鼠卵巢局部自分泌及旁分泌因子的调节作用。方法选用4月龄雌性大鼠(正常组),14月龄阴道细胞学表现动情期延长的雌性大鼠(作为老年大鼠模型)。模型大鼠随机分为宁心红杞大、中、小剂量组、阳性药(雌二醇)组和模型组,分别给予相应药物。HE染色观察卵巢生长卵泡、黄体等情况;免疫组化检测卵巢雌激素受体(ER)、孕激素受体(PR)、促卵泡激素(FSH)、促黄体激素(LH)、抑制素α(INHα)、激活素(ACT)α-β、卵泡抑制素(FS)、胰岛素样生长因子(IGF)-1的表达。结果与模型组比较,宁心红杞大、中剂量组卵巢指数、生长卵泡数增加(P<0.05),FSH、LH水平表达降低(P<0.05);宁心红杞大剂量组和阳性药物组ER表达水平上升(P<0.05),宁心红杞大、中剂量组IGF-1和INHα表达水平升高(P<0.05),而ACTα-β和FS表达水平下降(P<0.05)。结论宁心红杞胶囊可使卵巢局部自分泌、旁分泌系统调节因子相互协调平衡,进而改善卵巢功能。Objective To explore the regulatory effect and mechanism of Ningxin Hongqi Capsule on local ovarian autocrine and paracrine factors in peri-menopausal rats. Methods SD female rats aged 4 months were allocated in a normal control group （A） and those aged 14 months with vagino-cytologic figure of oestrus elongation were allocated in a senile female rat model group （B）. Rats in Group B were subdivided into 5 groups randomly as the B1, B2 and B3 subgroups treated respectively with high, moderate and low dose Ningxin Hongqi Capsule, the B4 subgroup treated with estradiol and the B5 subgroup untreated for control. Rats＇ ovaries were obtained at the end of the experiment for observing the conditions of ovarian growing follicles and corpus luteum by HE staining, determining expressions of ovarian estradiol receptor （ ER）, progesterone receptor （ PR）, follicle-stimulating hormone （FSH）, luteinizing hormone （LH）, inhibin α （INHα）, activin （ACT）α-β, follistatin （FS）, and insulinlike growth factor （IGF-1）. Results As compared with Group B5, the ovary index, number of growing follicle were higher and levels of FSH and LH were lower in Group B2 and B3, expression of ER was higher in Group B1 and 134, IGF-1 and INHet was higher in Group B2 and B3, and ACTα-β and FS were lower （ all P 〈 0. 05 ）. Conclusion Ningxin Hongqi Capsule could adjust and balance the local ovarian autocrine and paracrine factors to improve the ovarian function.
