摘  要:目的探讨β-酪啡肽-7(β-CM-7)在体外对人乳腺癌细胞(MCF-7)、人卵巢癌细胞(SKOV3)和人神经母细胞瘤细胞(SK—N—SH)增殖和凋亡的影响。方法用MTT测定法观察β-CM-7在体外对MCF-7、SKOV3、SK—N—SH细胞增殖的影响。用流式细胞仪测定β-CM-7在体外对MCF-7和SKOV3细胞凋亡的影响。结果β-CM-7作用于MCF-7细胞24和48h,对其增殖影响不明显(P〉0.05)。2.5×10^-6 -2.5×10^-2g/L的β-CM-7作用72h,能抑制MCF-7细胞增殖(P〈0.05或P〈0.01),抑制率为17.50%~32.08%。β-CM-7作用MCF-7细胞72h,在2.5×10^-7~2.5×10^-2g/L剂量下,诱导肿瘤细胞的凋亡(P〈0.05或P〈0.01),凋亡率为9.23%~29.41%,而在作用48h后,仅在2.5×10^-3g/L的高浓度下具有诱导肿瘤细胞的凋亡作用,凋亡率为10.12%。β-CM-7作用于SKOV3细胞24~72h,只有在高剂量(2.5×10^-2g/L和/或2.5×10^-3g/L)时,抑制肿瘤细胞增殖(P〈0.05),抑制率为10.50%~19.44%。β-CM-7作用SKOV、细胞48h和72h,在2.5×10^-3~2.5×10^-2g/L高剂量下能诱导肿瘤细胞的凋亡(P〈0.05或P〈0.01),凋亡率为8.72%~17.83%。β-CM-7对SK—N—SH肿瘤细胞增殖影响无显著性(P〉0.05)。结论β-CM-7能够抑制MCF-7和SKOV3细胞增殖,诱导细胞凋亡,作用结果与其剂量和作用时间有关。β-CM-7对SK—N—SH细胞的增殖无明显影响。Objective To study the effects of β-casomorphin-7 ( β-CM-7 ) on the proliferation and apoptosis of human breast cancer cell lines ( MCF-7 ), human ovarian cancer cell lines ( SKOV3 ) and human neuroblastoma cell lines (SK-N-SH) in vitro. Methods Inhibitions of β-CM-7 on MCF-7, SKOV3 and SK-N-SH cell proliferation, in vitro, were investigated by MTT colorimetric assay. Apoptosis percentages of MCF-7 and SKOV3 cells induced with β-CM-7, in vitro, were measured by flow cytometric assay (FCM). Results β-Casomorphin-7 had no significant effect on the proliferation of MCF-7 cell cultured in 24 h and 48 h, but β-CM-7 at 2.5 ×10^ -6- 2.5 ×10^ -2 g/L significant inhibited MCF-7 cell proliferation in 72 h ( P 〈 0. 05 or P 〈 0. 01 ), inhibitory rate of which were 17.50% - 32.08%. Cultured in 72 h, β-CM-7 at 2.5 ×10^ -7 - 2.5 × 10^-2 g/L notably induced MCF-7 apoptosis (P 〈0. 05 or P 〈0. 01 ), its apoptosis rate being 9. 23% -29. 41%. If cuhured in 48h, only β-CM-7 at high dose (2.5 ×10^-3 g/L) could induce MCF-7 apoptosis (P 〈0.05). For SKOV3, no matter what cultured at 24 h, 48 h or 72 h, β-CM-7 at high dose(2. 5× 10^-3 and/or 2. 5 × 10^-2 g/L) could inhibit cell proliferation and induce cell apoptosis (P 〈0.05 or P 〈 0. 01 ), their inhibitory and apoptosis rate were 10. 50% - 19.44% and 8.72% - 17.83%, respectively. However, it wasn't distinct for the influence of β-CM-7 on SK-N-SH cell proliferation. Conclusion β-CM-7 could inhibit proliferation and induce apoptosis in MCF-7 and SKOV3, the effects depend on β-CM-7 dose and test time. β-CM-7 affects SK-N-SH very little.
