利用蜘蛛酯酶检测蔬菜中的有机磷农药残留

摘  要:探讨利用大腹园蛛酯酶检测蔬菜中有机磷农药残留的技术和方法.结果表明:大腹园蛛腹部的酯酶活性显著高于头胸部,在37℃和缓冲液pH为7.0时,酯酶活性最高.有机磷农药乐果浓度的对数值与酶抑制率存在较好的直线关系,通过检测蔬菜洗脱液对蜘蛛酯酶抑制率的方法可以用来检测蔬菜中的有机磷农药残留.采用该方法检测时,乐果的检出限为0.5094 mg/L.受检3种蔬菜的有机磷农药残留量均低于规定的标准.A technique for detection of organophosphorous(Ops) pesticide residues was explored by measuring the activities of esterase from Araneus ventricosus. The results were showed that the esterase activities in abdomen from the adult female spider were higher than that in cephalothorax. The esterase activity was the highest under 37℃ and with pH 7. 0 of phosphate buffer solution. There was a significant liner regression between the concentration logarithm of dimethoate and the inhibitory rate of esterase. The Ops pesticide residues can be detected by measuring the inhibitory rate of vegetables washing liquid to the esterase from the spider. The detection limit of dimethoate was 0. 509 4 mg/L by this method. The Ops pesticide residues in three species of vegetables were lower than the standard values given by Chinese government and FAO/WHO.
