针药并用对大鼠子宫内膜异位症组织中基质金属蛋白酶-2表达的影响

摘　　要：目的探讨针灸配合中药加味没竭片对子宫内膜异位症(EMs)大鼠基质金属蛋白酶-2(MMP-2)、MMP-2 mRNA表达的影响。方法建立大鼠EMs模型,随机分为模型组、针灸组、中药组、针药组,并设立假手术组。针灸组电针"血海"、"三阴交",艾灸"关元";中药组采用加味没竭片生理盐水混悬液灌胃;针药组采用上述两种方法综合治疗;假手术组和模型组捆绑固定并行生理盐水灌胃。治疗35天后,测量各组大鼠异位组织长、宽、高三径,免疫组化法检测组织中MMP-2的表达,荧光定量聚合酶链反应法检测组织中MMP-2 mRNA的表达。结果针药组异位组织大小组织中MMP-2的表达均明显低于模型组、针灸组和中药组,MMP-2 mRNA的表达水平各组之间差异没有统计学意义(χ2=0.508,P=0.917),异位组织中MMP-2蛋白表达与MMP-2 mRNA表达间没有直接的线性相关性(r=0.117,P=0.473)。结论针药并用治疗能缩小EMs大鼠子宫内膜异位组织大小,下调异位组织中异常升高的MMP-2蛋白表达,但对异位组织中MMP-2 mRNA的表达影响不大。Objective To explore the effect of acupuncture and moxibustion plus Jiawei Mojie Tablet on the expressions of matrix metalloproteinase-2 （MMP-2） and MMP-2 mRNA in rats with endometriosis （EMs）. Methods A rat model of EMs was made. The rats were randomly divided into model, acupuncture and moxibustion, Chinese herbal medicine, and combined acupuncture and medi- cine groups. A sham operation group was also set up. The acupuncture and moxibustion group was treated by electroacupuncture of Xuehai（SP 10） and Sanyinjiao（SP 6） and moxibustion of Guanyuan（CV 4）. The Chinese herbal medicine group was treated by gavage of Jiawei Mojie Table suspension in normal saline. The combined acupuncture and medicine group was treated by a combination of the above two methods. The sham operation and model groups were bound, fixed and gavaged with normal saline. After 35 days＇ treatment, the length, width and height of the endometrial explant were measured; the expressions of MMP-2 and MMP-2 mRNA in the explant were examined by immunohistochemistry and fluorescence quantitative polymerase chain reaction, respectively, in each group of rats. Results The largest diameter of the explant and the expression of MMP-2 in the explant were significantly lower in the combined acupuncture and medicine group than in the model, acupuncture and moxibustion, and Chinese herbal medicine groups. There was no statistically significant difference in the expression of MMP-2 mRNA between the various groups （x^2 =0. 508, P =0.917）. There was no direct linear correlation between the expressions of MMP-2 and MMP-2 mRNA in the explant （r =0. 117, P = 0. 473）. Conclusion A combination of acupuncture and medicine can reduce the volume of the endometrial explant in endometriosis rats and down-regulate abnormally increased expression of MMP-2 in the explant but does not have an obvious influence on MMP-2 mRNA expression.
