摘 要:目的观察溃结灵对溃疡性结肠炎(UC)大鼠结肠黏膜Toll样受体2(TLR2)的影响。方法采用三硝基苯磺酸(TNBS)法制作UC大鼠模型,将大鼠随机分为正常对照组、模型组、溃结灵低、中、高剂量组和阳性药柳氮磺胺吡啶(SASP)组。治疗10d后脱颈椎处死大鼠,取新鲜结肠黏膜标本并提取全细胞蛋白,采用蛋白免疫印迹(Western blot)方法对TLR2的蛋白表达水平进行检测,以β-actin作为内参,以TLR2蛋白与β-actin密度的比值作为目的蛋白的相对含量。结果模型组TLR2蛋白相对表达量明显高于正常组(P<0.01);溃结灵中、高剂量组和SASP组TLR2相对表达量明显低于模型组(P<0.05或P<0.01)。结论TLR2可能参与了UC大鼠的发病过程,溃结灵使TNBS法UC大鼠结肠黏膜TLR2的蛋白表达水平明显降低,这可能是其治疗UC的机理之一。Objective To explore the changes of proteins level of TLR2 in colonic mucosa of ulcerative colitis (UC) model rats and to observe the effects of Kuijieling Decoction (KD) on TLR2. Methods UC model rats were induced by TNBS. The rats were randomly divided into six groups as follows: normal control (NC) group, model control (MC) group, low dose Kuijieling (KLD) group, medium - dose Kuijieling (KMD) group, high - dose Kuijieling (KHD) group and SASP group. After 10 - days treatment the rats were killed to get their colonic mucosa and extract the whole - cell proteins. Western blot technique was used to detect the TLR2 protein level. β - actin was used as the internal index. Relative expression level of target protein was calculated from the gray scale ratio of TLR2 and β- actin. Results Relative protein expression level of TLR2 in MC group was significantly higher than that in NC group ( P 〈 0.01 ). Relative protein expression of TLR2 in KMD and KHD group was significantly lower than that in MC group ( P 〈 0. 05 or P 〈 0. 01 ). Conclusion TLR2 may be involved in the pathogenesis of UC, and KD could inhibit relative protein expression of TLR2 in colonic mucosa of UC model rats induced by TNBS. The inhibitory effects of KD on UC might be associated with the inhibition of protein expression of TLR2.
