健胃愈疡颗粒对胃溃疡大鼠胃黏膜的保护机制

摘　　要：目的探讨健胃愈疡颗粒对胃溃疡大鼠胃黏膜的保护作用及其可能机制。方法用Okabe改良法复制大鼠胃溃疡模型,观察健胃愈疡颗粒对溃疡指数、胃黏膜组织形态的影响,并以免疫组织化学检测IL-1β和bFGF蛋白的表达水平。结果健胃愈疡组溃疡指数明显低于模型组和法莫替丁组;HE染色法莫替丁和健胃愈疡组较模型组黏膜厚度增加,溃疡床及溃疡周围炎性细胞浸润明显减少,纤维排列较整齐;正常组及假手术组胃组织仅见微量的IL-1β蛋白表达,模型组、健胃愈疡组、法莫替丁组表达均增强,以模型组最明显(P<0.01);健胃愈疡组与法莫替丁组间差异无显著性;bFGF蛋白在正常及假手术组呈弱阳性表达;术后除正常及假手术组外,各组大鼠胃黏膜bFGF蛋白表达增强(P<0.01);健胃愈疡组、法莫替丁组均高于模型组(P<0.01);健胃愈疡组与法莫替丁组比较差异无显著性。结论健胃愈疡颗粒能保护大鼠胃黏膜组织,预防乙酸诱导性胃溃疡,降低造模大鼠溃疡指数;抑制大鼠胃黏膜IL-1β蛋白的表达,增加bFGF蛋白的表达,促进溃疡愈合。Objective To investigate the protective effects and its mechanism of Jianwei Yuyang Granule （JYG） on gastric mucosa of experimental gastric ulcer rats. Methods The model of gastric ulcer was established by the modified Okabe＇ s method. The ulcer index was recorded, and the expression of IL - 1β and bFGF was determined with histochemistry. Results The ulcer index of JYG group was significantly lower than that of model group and famotidine group （ P 〈 0. 01 ）. The expression of IL - 1β was at an extremely lower level in normal mucosa, but was obviously higher in model group than in sham - operation group, JYG group and famotidine group（ P 〈 0. 01 ）. And no significant difference of IL - 1β expression was seen between JYG group and famotidine group. On the other hand, the expression of bFGF was at low level in normal mucosa and sham - operated rats and increased in the JYG group, model group and famotidine group, and the level in JYG group and famotidine group was higher than that in the model group（ P 〈 0. 01 ）. Conclusions JYG can prevent the gastric mucosa of rats from suffering gastric ulcer induced by acetic acid, and the increase of bFGF ex- pression and the reduction of IL - 1β expression may be its proactive mechanism.
