摘  要:为了选出一种适合水稻纹枯病菌RNA的提取方法,比较了改良的盐酸胍法同热酚法、Trizol法提取水稻纹枯病菌RNA的效果。结果表明:3种方法中,改良的盐酸胍法和Trizol法可以提取到高质量的RNA,热酚法的提取效果不理想。改良的盐酸胍法提取的RNA进行mRNA差异显示的结果表明,没有多糖对后继反应的干扰,是一种经济有效的水稻纹枯病菌RNA的提取方法。用该法提取RNA,可以满足分子克隆与基因表达分析等分子生物学后继试验。To select a method for Rhizoctonia solani Kuhn RNA extraction.The improved Guanidine HCl method,hot Phenol method and the Trizol method were applied to isolate total RNA from Rhizoctonia solani Kuhn.In the three methods,the improved Guanidine HCl method and the Trizol method could prepare high quality and quantity RNA while the result of the hot phenol method was not good.The mRNA differential display results showed that there was no polysaccharides influenced the consequent reaction in the RNA isolated by the improved Guanidine HCl method.The improved Guanidine HCl method was an economic and efficient method for preparation of Rhizoctonia solani Kuhn RNA.The RNA prepared by this method could meet the needs of most molecular biological experiments including gene cloning and expression analysis.
