摘  要:自身免疫性甲状腺疾病(AITD)的特征是甲状腺组织中存在大量T细胞浸润。对T细胞表面的抗原识别受体(TCR)α、β链可变区进行基因谱检测,对于分析与确定特异性自身反应性T细胞克隆具有重要意义。研究发现,无论是AITD还是甲状腺炎动物模型,甲状腺组织内浸润的T细胞TCRα、β可变区的某些特定亚家族不同程度选择性高表达。以表达特定TCR亚家族的特异性T细胞为靶抗原,基于TCR的单克隆抗体、多肽疫苗和DNA疫苗可能是治疗AITD的有效手段。The human autoimmune thyroid disease (AITD) is characterized by profuse infiltrating of both CD4^ + and CD8^+ T cells. Examination of the T cell receptor (TCR) variable (V)gene repertoire is important in the analysis of the autoimmune reactive T cells clone in AITD. The intrathyroidal TCR repertoire in human AITD and animal models of induced and spontaneous autoimmune thyroiditis has been shown to be biased as evidenced by the use of a restricted TCR V gene repertoire seen in both TCR alpha and beta chains. The restricted distribution and clonal expansion of TCR V alpha or V beta subfamily T cells permit the identification of specific antigen responses of T cells. Immunopathologically, T cells that respond to thyroid-specific autoantigens play important roles in the development of lesions in the target organ. One potential mode of treatment would be the target of effector lymphocytes bearing that particular TCR. Therefore, TCR-based monoclonal antibodies, peptide vaccines and DNA vaccines can be used for treatment of AITD.
