摘  要:目的探讨亚低温对缺氧缺血脑损伤(HIBD)新生大鼠脑源性神经营养因子(BDNF)mRNA表达的影响。方法新生7日龄Wistar大鼠84只,随机分为:假手术组(n=6),31℃亚低温干预组(n=36),非亚低温干预组(对照组,n=36)及正常对照组(n=6)。于分组后6、12、24、48、72、96h用原位杂交技术测定BDNFmRNA。用Tunel染色和免疫组化技术检测神经元凋亡数。结果HIBD缺血再灌注后脑组织BDNFmRNA表达增加,亚低温组HIBD脑组织BDNFmRNA表达与未干预组比较差异有统计学意义,亚低温组脑组织海马、皮层区细胞凋亡数与未干预组比较差异有统计学意义。结论亚低温可通过增加HIBD后脑组织海马、皮层BDNFmRNA的表达水平,抑制细胞凋亡而发挥脑保护作用。Objective To study the effect of mild hypothermia on brain-derived neurotrophic factor (BDNF) mRNA in neonatal rats with hypoxic-ischemic brain damage (HIBD). Methods Eight-four Wistar rats selected for this study that were 7 days old and weighed 13 to 16 g, The rats were randomly assigned into four groups, control group ( n = 6), sham operated group ( n = 6 ), HIBD group ( n = 36) and mild hypothermic group (n = 36). The expression of BDNF mRNA of brain tissue was measured at 6, 12, 24, 48, 72 and 96 h by in situ hybridization. The apoptosis of neurons was measured by Tunel and HE kit. Results It was found that there was an increase of expression of BDNF mRNA in the dentate gyrus and hippocamapal CA1 area after hypoxic-ischemic injury in rats. It was significantly higher in level of BDNF mRNA of rats in mild hypothermic group than that in HIBD group. There was significant difference of apoptosis in neurons in gyrus and hippocamapal CA1 area of rats in mild hypothermia group compared with that in HIBD group. Conclusion Mild hypothermia can protect brain tissue by increasing the level of expression of BDNF mRNA and inhibiting apoptosis of neurons.
