华蟾素对卵巢癌3AO细胞MMP-2和TIMP-2表达的影响和意义

摘　　要：目的：探讨华蟾素对卵巢癌3AO细胞MMP-2和TIMP-2表达的影响及临床意义。方法：体外培养卵巢癌3AO细胞，不同浓度的华蟾素干预后，应用SABC免疫组化法检测MMP-2和TIMP-2的表达。结果：体外培养的卵巢癌3AO细胞可表达MMP-2和TIMP-2；0．25mg／ml华蟾素对卵巢癌3AO细胞MMP-2和TIMP-2的表达无显著影响（P〉0．05）；2．5mg／ml华蟾素干预后，卵巢癌3AO细胞MMP-2的表达显著下降，TIMP-2的表达显著升高（P〈0．05）；25mg／ml华蟾素与2．5mg／ml华蟾素对卵巢癌3AO细胞MMP-2和TIMP-2表达的影响无统计学差异（P〉0．05）。结论：2．5mg／ml华蟾素可显著降调卵巢癌3AO细胞MMP-2的表达，升调TIMP-2的表达，MMP-2和TIMP-2表达的失衡可抑制卵巢癌3AO细胞的侵蚀性生长。Objective： To investigate the effect and clinical significance of cinobufacine on the expression of MMP - 2 and TIMP - 2 in the ovarian carcinoma 3AO cells cultured in vitro. Methods：Ovarian carcinoma 3AO cells were cultured in vitro ;after the action of cinobufacine with different concentration, the expression of MMP- 2 and TIMP -2 was measured by SABC immunohistometry technique. Results：The ovarian carcinoma 3AO cells cultured in vitro could express MMP -2 and TIMP -2 ;0. 25mg/ml cinobufacine didn＇t affect the expression of MMP -2 and TIMP -2 in the ovarian carcinoma 3AO cells（ P 〉0.05 ） ;after the action of 2. 5mg/ml cinobufacine, the expression of MMP- 2 was down regulated and the TIMP - 2 was unregulated significantly （ P 〈 0. 05 ） ; the action of 25mg/ml cinobufacine on the MMP - 2 and TIMP - 2 was similar to that of 0. 25mg/ml cinobufacine（P 〉0. 05 ）. Conclusion：2. 5mg/ml cinobufacine can down regulate the expression of MMP - 2 and up regulate the expression of TIMP -2. The imbalance of MMP -2 and TIMP -2 may inhibit the invasive growth of ovarian carcinoma 3AO cells.
