摘  要:目的观察蒲地灌肠液灌胃给药对小鼠的急性毒性反应及对家兔直肠粘膜的刺激性。确定安全有效的给药剂量,确保患者用药安全。方法将蒲地灌肠液浓缩后,分四个剂量组,给小鼠灌胃给药,观察给药后的毒性反应。按20ml.kg-1剂量,每日1次对家兔灌肠给药,与生理盐水组比较,观察给药组的直肠粘膜刺激性。结果急性毒性实验,在剂量为66.2g.kg-1(相当于原药液105ml.kg-1)时,所有给药小鼠均未发生中毒反应或死亡;刺激性试验,与对照组相比,无显著差异。结论蒲地灌肠液小鼠急性试验最大耐受量为66.2g.kg-1,是人用灌肠量的63倍,可认为该药在正常灌肠剂量下无急性毒性,对家兔直肠粘膜无刺激性。OBJECTIVE To investigate the stimulation and the acute toxicity of Pudi enema.To determine a safe and effective medicine dosage,ensure the patient to use the medicine security.METHODSThe toxic responses of Pudi enema were observated by intragastric administration to mice.The rectal mucosa irritation were observated by rectum administration to rabbits.RESULTSWhen the dosage was 66.2g·kg-1(quite Pudi enema 105ml·kg-1),all the tested mice showed no toxic responses.There was no significant difference in retal mucous membrane of tested rabbits compared with controled rabbits.CONCLUSIONAcute toxicity test showed that maximun tolerance was 66.2g·kg-1,which was 63 times of the amount commonly used in clinic.Our work suggested that Pudi enema had no obvious acute toxicity and had no stimulative response on mucous membrane.
