乙型脑炎病毒HW_1克隆纯化株的鉴定及外源性污染的检定

摘　　要：应用病毒蚀斑技术，将乙型脑炎病毒（ＪＥＶ）ＨＷ１株在鸡胚成纤维细胞（ＣＥＦ）单层培养上连续挑斑纯化３次，对纯化后的克隆株进行血清学、免疫生物学鉴定及外源性污染检定。毒株对ＢＨＫ－２１细胞感染滴度≥１０６．１５ＴＣＩＤ５０；对８～９ｇ小鼠脑内感染的毒力≥１０７．１０ＬＤ５０／０．０４ｍＬ；与ＪＥＶＰ３株阳性参考血清的中和指数≥２０００；克隆株对猪有较好的免疫原性，无外源性污染。Japanese B encephalitis virus HW 1 strain was plague purified three times in chick embryo fibroblast (CEF). The purified strain was identified with serological and immunobiological technics and its outer contamination was detected. The titer of the strain against BHK 21 cells was more than 10 6.15 TCID 50 . The virulence for the mice (weight of 8 9 g) to be infected via encephalon was more than 10 7.10 LD 50 /0.04 mL. The PD 50 of the strain with positive reference serum of JEV P 3 strain was more than 2 000. The strain was proved to be a good immunogen to swine and have no outer contamination.
