摘 要:【目的】确定GAS6(growt harrested-special 6)基因在染色体的物理位置,为该基因的克隆及功能研究打下基础。【方法】运用比较基因学的方法,根据人的该基因序列设计引物,从大围子猪基因组分离猪该基因的内含子9的片段,通过扩增体细胞杂种克隆板上27个样品和辐射克隆板上118个样品。【结果】分离了猪该基因包括内含子9的片段(GenBank收录号为AY880668),首次将GAS6基因物理定位于猪SSC11q11-17,与微卫星SW1452标记紧密连锁,LOD值为16.88,存留率为22%,在放射杂交图谱上的图距为56cR。【结论】GAS6基因区间定位结果与精细定位结果相一致,也与比较定位结果相一致,并进一步验证了猪11号染色体(SSC11)和人大部分13号染色体(HSA13)存在同源性。【Objective】The objective of this study is to map the gene of GAS6 in the position of chromosome in pigs and to lay a foundation of cloning of GAS6 gene. 【Method】The pig × rodent somatic cell hybrid panel (SCHP) and INRA-Minnesota porcine radiation hybrid panel (IMpRH) were used. 【Result】The isolated GAS6 gene of swine was deposited into GenBank and the accession number was AY880668. The results showed that the gene of GAS6 was located on the SSC11q11-17. The GAS6 gene was linkaged closely with the microsatellite SW1452 anchored on SSC11 with the rate of retension of 0.22 and LOD score threshold was 16.88 and the distance in map of the radiation hybrid (RH) was 56 cR. 【Conclusion】The position of GAS6 gene in SSC11 by means of IMpRH conformed to the conclusions that the SSC11 is homologous with most part of HSA11.
