摘  要:[目的]研究丙硫氧嘧啶(Propylthiouracil,PTU)干扰甲状腺功能的亚急性毒性状况,为PTU致甲状腺肿的组织发生学研究提供实验室基础数据。[方法]SD大鼠150只,随机分为15组,包括3、6、9、12、15、18、21d的试验组和同期对照组,对照组用玉米油灌胃,试验组用PTU的玉米油溶液灌胃,于每个实验期结束的d2麻醉后股动脉放血处死动物,取甲状腺、肝、肾、脾、胸腺,称重后进行组织病理学观察。[结果]试验组血清TT3、TT4浓度先降低再进入平缓期,TSH浓度是先升高再进入平缓期,甲状腺绝对重量和脏器系数均持续升高,和对照组相比,6d时差异有统计学意义(P﹤0.05)。[结论]PTU5mg/(kg.day)的剂量染毒大鼠21d,除干扰甲状腺功能,未见其他毒性反应,甲状腺在持续增生,功能尚未代偿。因此用该方法研究甲状腺的组织发生学是可行的。[Objective]To study the subacute toxicity of PTU on thyroid function in rats,so as to provides basic information of laboratory for the study of goiter histogenesis induced by PTU.[Methods]150 SD male rats were randomly divided into 15 groups including intervention groups and control groups by 2d,6d,9d,12d,15d,18d,and 21d. These rats were sacrificed by blood letting through arteria femoralis at the day after the end of each experiment period. The thyroid,liver,kidney,spleen and thymus were weighted and observed the change of histopathology.[Results]The concentration of TT3、TT4 in serum of experiment group decreased and then showed to be plat. The concentration of TSH firstly increased then kept stable,and weight of thyroid and coefficient of organ were keep in a high level,which showed significant difference at 6 day when compared with control group(P﹤0.05).[Conclusion]When the dosage of PTU was 5mg/(kg.day)for 21 days,the function of thyroid was disrupted and thyroid continued to proliferation,while not any other positive effect was observed as well as the compensation of its function. So this method is feasible to research on the histogenesis of thyreoid.
