实验性氟中毒大鼠大脑皮质c-fos和Caspase8表达的观察

摘  要:目的探讨慢性氟中毒对大鼠大脑皮层神经元的影响,为慢性氟中毒对中枢神经系统的损害提供形态学依据。方法采用40日龄Wistar雄性大鼠,以喂饲高氟(100mg/L)水的方法建立慢性氟中毒模型。对其大脑皮层神经元进行c-fos免疫组织化学染色和半胱氨酸天冬氨酸蛋白酶8(Caspase8)原位杂交反应。结果c-fos免疫组织化学分析,对照组阳性细胞率为32.1%,实验组阳性细胞率为35.8%,两组比较差异无统计学意义(χ^2=0.305,P〉0.05);显微图像定量分析,对照组灰度值为0.2774±0.033l,实验组灰度值为0.2756±0.0241。两组比较差异无统计学意义(χ^2=0.826,P〉0.05)。Caspase8原位杂交反应,对照组阳性细胞率为14.1%,实验组阳性细胞率为18.7%,两组比较差异无统计学意义(χ^2=0.419,P〉0.05);显微图像定量分析,对照组灰度值为0.3884±0.0323,实验组灰度值为0.3874±0.0329,两组比较差异无统计学意义(χ^2=0.641,P〉0.05)。结论慢性氟中毒对大鼠大脑皮层神经元发生凋亡的影响不明显。Objective To explore the effects of chronic fluorosis on neurons in the cerebral cortex of rats, and to provide some morphological evidence of damage in the central nervous system induced by chronic fluorosis. Methods Male Wistar rats 40 days after birth were fed with high fluoride contented water(100 rag/L) for inducing chronic fluorosis. Immunocytochemistry and in situ hybridization were used to detect c-fos and Caspase 8 at cerebral cortical neurons respectively. Results c-fos positive cells rate and gray scale in the cerebral cortex of chronic fluorosis were 35.8% and 0.2756 ± 0.0241, respectively, and that of control group were 32.1% and 0.2774 ±0.0331 with statistical difference(χ^2= 0.305, t = 0.826, P〉 0.05). Caspase 8 positive cells rates of fluorosis group and control group were 18.7% and 14.1%, respectively, the difference being statistically significant(χ^2= 0.419, P 〉 0.05). The gray scale of fluorosis group and control group were 0.3874 ± 0.0329 and 0.3884 ± 0.0323, respectively, the difference being statistically significant(t = 0.641, P〉 0.05). Conclusion Chronic fluorosis had no significant influence on apoptosis of cerebral cortical neurons.
