摘 要:目的观察新生大鼠缺氧缺血性脑损伤(HIBD)后脑内神经蛋白聚糖的动态变化,探讨神经可塑性相关抑制因素在发育期脑损伤后的改变。方法7d龄SD大鼠随机分为假手术组和HIBD组,于脑缺氧缺血术后1,4,7,14,28d分别取脑组织进行H-E染色和免疫组织化学染色观察组织病理变化及神经蛋白聚糖蛋白表达,并以RT-PCR法检测其mRNA表达。结果假手术组神经蛋白聚糖的表达于术后7d达到高峰(P<0.05),此后阳性表达逐渐减少;HIBD组术后4d、7d的神经蛋白聚糖蛋白及其mRNA表达均低于假手术组(P<0.05),至术后14d、28d表达增高,均高于假手术组(P<0.05)。结论新生大鼠HIBD后脑组织神经蛋白聚糖蛋白及其mRNA表达随着时间推移发生变化,提示HIBD后神经可塑性调控的关键时期为损伤后2周内。Objective To observe the change of neurocan in neonatal rat brains after hypoxiaischemia brain damage(HIBD), and explore the change of inhibitory factors neuronal plasticity associated with neonatal brain damage. Methods 7 days neonate SD rats were randomly divided into sham-operated group and HIBD group. Then at 1, 4, 7, 14, 28 days after the operation, dissect the brain and the expression of neurocan was detected by reverse transcription-polymerase chain reaction(RT-PCR) and immunohistochemical method respectively. Results The amount of neurocan reached a peak level after 7 days of the operation in sham-operated brains (P〈0.05), then reduced. Expression of neurocan in HIBD brains at 4, 7 days after the operation was lower than that of sham-operated brains(P〈0.05), while was higher than that of sham-operated brains at 14, 28 days after the operation(P〈0.05). Conclusion The expression of neurocan is different in the progress of neonatal HIBD and the best time for controlling neuronal plasticity associated inhibitory factors is within two weeks.
