摘 要:目的:研究癣湿涂膜剂的制备工艺并对其透皮吸收性能进行初步评价。方法:以蛇床子素和总生物碱的含量为指标,采用正交实验筛选提取工艺;从成膜性能、稳定性、溶解性等方面优选涂膜剂的成膜材料;用直立式扩散池进行离体小鼠皮体外释放试验,HPLC法测定蛇床子素4 h的累积释放量。结果:优选的最佳提取工艺为:药材粉碎后,以6倍量70%的乙醇为溶剂渗漉;涂膜剂的成型以7%的PVA-1788最佳;涂膜剂4 h的透皮率明显高于原始液体制剂。结论:改进后的药材提取工艺具有更高的提取率,涂膜剂制备工艺简单可行,透皮吸收有效,可作为代替原剂型的新方法。Objective: To prepare the Xuanshi Liniment and evaluate its quality of transdermal absorption. Methods: Optimized extracting method of Xuanshi Liniment was determined by using the orthogonal experiment. Best film material was selected based on capability, stablity and solubility. In vitro permeation behavior was studied in vertical diffusion cell. Samples were assayed by the HPLC method. Results:The 4 h cumulative permeated rate of osthole from Xuanshi Liniment through mouse skin was much higher than that from original lotion. Conclusion: The Xuanshi Liniment is more effective in transdermal absorption with high extraction, simple and feasible preparation, and may be supersession of the original lotion.
