滇产粗根荨麻水提取部分对佐剂性关节炎大鼠腹腔巨噬细胞分泌TNF-α的影响

摘　　要：目的:探讨滇产粗根荨麻水提取部分对佐剂性关节炎(adjuvant arthritis,AA)大鼠足趾肿胀及腹腔巨噬细胞(peritoneal macrophages,PMф)分泌肿瘤坏死因子-α(tumor necrosis factor-alpha,TNF-α)的影响。方法:建立大鼠佐剂性关节炎模型,脂多糖(lipopolysaccharide,LPS)诱导大鼠腹腔巨噬细胞分泌TNF-α,用酶联免疫吸附法检测。结果:滇产粗根荨麻水提取部分(400mg/kg,200mg/kg)能明显抑制佐剂性关节炎大鼠的足肿胀,并降低大鼠腹腔巨噬细胞分泌TNF-α水平。结论:滇产粗根荨麻水提取部分对佐剂性关节炎的治疗作用可能与其抑制腹腔巨噬细胞分泌TNF-α有关。Objective ： To explore the effects of the aqueous fraction from Urtica nuwrrhiza Hand-Mazz on the level of TNF-α from peritoneal macrophages （PMФ） in adjuvant arthritis rats. Methods： Freund＇s complete adjuvant was injected in the hind paw of rats to induce adjuvant arthritis （AA） rat model. The edema of the hind paw was measured at different time. The level of TNF - α from PMФ was measured by ELISA method. Results： Compared with adjuvant arthritis group, the aqueous fraction from Urtica macrorrhiza Hand-Mazz at a dose range of 200mg to 400mg can significantly relieve the paw edema and inhibit LPS induced release of TNF - α from PMФ in AA rats. Conclusion： Aqueous Fraction from Urtica macrrhlza Hand-Mazz was found to inhibit the production of TNF - α, adjuvant arthritis rats, it might be related to the mechanisms of its anti-inflammation.
