CD40单克隆抗体与前房相关免疫偏离小鼠中脾脏CD8^＋T细胞关系的初步研究

摘　　要：目的:OVA小鼠前房抗原特异性CD8+T细胞的数量和功能变化是否与CD40单克隆抗体有关。方法:OVA前房注射的同时,腹腔给与小鼠CD40单克隆抗体,第7d给予足底皮下免疫OVA和CFA,第14d,取脾脏制备单细胞悬液,用流式检测和细胞内细胞因子染色(IFN-γ)进行评价CD8+T细胞的数量和功能有无变化。同时用不给与ipCD40单克隆抗体的ACAID小鼠作为阴性对照组。结果:试验组和阴性对照组脾脏CD8+T细胞数量和分泌IFN-γ无显著性差异。结论:ACAID中,CD8+T细胞介导的细胞毒性反应(CTL反应)可能为CD4+T细胞非依赖性。AIM： To evaluate the the relationship between CEH0 mAb and CD8^＋ T cells in the spleen of anterior chamber associated immune deviation （ACAID） mice. METHODS： The mice were injected into anterior chamber with ovabumin （OVA） and given intraperitoneal immunization CD40 mAb at the same time in experimental group. On day 7, these mice were immunized subcutaneously with antigens and complete Freund＇s adjuvant （CFA）. On day 14, the spleen was extracted and the single cell suspension was made. With flow cytometry （FCM） and intracellular IFN-γ staining, the number and function of CD8^＋ T cells were reevaluated. The ACAID mice without intraperitoneal immunization CE40mAb were used as negative control groups. RESULTS： There was no significant difference in the number of CD8^＋ T cells and IFN-γ secretion between experimental group and negative control group. CONCLUSION： The CTL reaction in ACAID might be CD4^＋ T cells independent.
