固定不同时间对兔骨骼肌肌动蛋白免疫组化染色影响的研究

摘  要:目的探讨标本固定不同时间对骨骼肌肌动蛋白免疫组化染色的影响。方法采用免疫组织化学SABC法和图像分析技术检测兔死后骨骼肌经福尔马林液固定不同时间(12h、1d、2d、4d、8d、16d、24d、32d)肌动蛋白免疫组化染色的变化。结果骨骼肌组织固定32d内,镜下观察各组肌动蛋白免疫组化染色结果未见明显变化,经图像分析检测和统计分析结果表明:各组之间肌动蛋白表达的R值和OD值的差异无统计学意义。结论骨骼肌组织固定32d内,对肌动蛋白免疫组化染色效果无明显影响。Objective To study the influence of fixing time of samples on actin expression in the skeletal muscle of rabbits by immunohistochemical staining. Methods The changes in actin expression of skeletal muscle at different fixing times (12h,ld ,2d ,4d,8d, 16d,24d or 32d) were observed microscopically by immunohistochemical staining (SABC method) and analyzed by image analysis system. Results The specimens of normal skeletal muscle fixed within 32 days showed homogeneous brown reaction for actin, and there were no obvious changes at the different fixing times. The result of statistical analysis also indicated that the R and OD values of actin expression had no significant difference among the different groups. Conclusion There is no obvious difference in actin staining in skeletal muscle fixed within 32 days.
