我国山丘、平原疫区利什曼原虫分离株kDNA分析

摘  要:为鉴别我国山丘、平原疫区杜氏利什曼原虫分离株,采用内切酶AluⅠ对其kDNA进行酶切。结果显示实验各株均有500bp片段;L.d.汶川人株、L.d.四川人株、L.d.甘肃人株(GS2)、L.d.四川犬株、L.infantum及L.d.Jeddah具有210bp片段;L.d.四川人株、Ld.甘肃人株(GS2)及L.d.四川犬株具有120bp片段;L.d.汶川人株、L.d.甘肃人株(GS2)和L.d.四川犬株具有400bp和280bp片段。L.d.甘肃犬株也具有400bp片段;L.infantum和L.d.Jeddah具有280bp片段。提示山丘疫区不同分离株的kDNAAluⅠ酶切片段相近,其中人株与犬株的相似;山丘疫区分离株与平原疫区分离株的酶切片段之间有较大差异,山丘疫区分离株与L.infantum的酶切片段之间既有相似又有差异。To identify Leishmania donovani isolates from hill and plain foci of China, and to reveal the intrinsic relation between them and L. infantum, the kDNAs of Leishmania isolates from different foci were digested by Alu I endonuclease.The results showed that the 500bp fragment existed in all the isolates observed. 210bp fragment existed in L. d. Wenchuan human isolate, L.d. Sichuan human isolate, L. d. Gansu human isolate (GS2),L. d. Sichuan canine isolate, L. infantum and L. d .Jeddah; 120bp fragment existed in L. d. Sichuna human isolate, L. d. Gansu human isolate (GS2) and L. d. Sichuan canine isolate; 400bp and 280bp fragments existed in L. d. Wenchuan human isolate, L. d. Gansu human isolate (GS2) and L. d. Sichuan canine isolate, 400bp fragment also existed in L. d. Gansu canine isolate; 280bp fragment existed in L. infantum and L.d. Jeddah. It is suggested that the Alu I digested kDNA fragments within isolates fiom hill foci are similar, in which the fragments of human isiolates are similar to that of canine isolates; but the fragments between isolates from hill foci and plain foci are quite different, and which between isolates from hill foci and L. infantum are both similar and differen.
