摘 要:目的:探讨天津地区人群中过氧化物酶体增殖物激活受体γ(PPARγ)基因Pro12Ala及toll样受体4(TLR4)基因Asp299Gly、Thr399Ile的多态性与2型糖尿病(T2DM)的关系。方法:提取365例T2DM患者(T2DM组)和95例健康人(对照组)的基因组DNA,PCR扩增目的片段后,用变性高效液相色谱技术(DHPLC)筛查基因突变情况。结果:T2DM组和对照组PPARγ基因Pro12Ala位点P/A的基因型频率分别为0.0740和0.0316(27/365和3/95),差异无统计学意义(P>0.05);两组均未筛查到TLR4基因Asp299Gly、Thr399Ile突变基因型。结论:天津地区人群PPARγ基因Pro12Ala、TLR4基因Asp299Gly、Thr399Ile多态性与T2DM无明显关联。Objective: To explore the relationship among peroxisome proliferator-activated receptorγ (PPARγ) gene Pro12Ala and toll-like receptor 4(TLR4) gene Asp299Gly, Thr399Ile polymorphisms and type 2 diabetes mellitus (T2DM) in Tianjin area. Methods: To extract the genome DNA from 365 patients with T2DM and 95 healthy controls,and use polymerase chain reaction (PCR) to amply the objective fragment,then use DHPLC to detect the mutation. Results: There were no significant differences in genotype frequencies (P/A) of PPARγ gene Prol2Ala polymorphism between T2DM and control groups (0.074 0,27/365 and 0.031 6,3/95, respectively,P 〉 0.05 ). The genotypes of TLR4 gene Asp299Gly and Thr399Ile mutations were not found in the two groups. Conclusion: There were no significant correlation among PPARγgene Prol2Ala,TLR4 gene Asp299Gly,Thr399Ile polymorphisms and type 2 diabetes mellitus in Tianjin area.
