摘 要:目的:研究参麦注射液对脑出血大鼠血肿周围区缺氧诱导因子1-α(Hypoxia inducible factor 1-α,HIF1-α)表达的影响及其意义。方法:将50只SD大鼠随机分为正常组、假手术组、模型组、参麦组4组,模型组和参麦组组内又分为1、3、5、7天共4个时间点,立体定位仪定位注射胶原酶VII造大鼠脑出血模型,观察大鼠脑出血后神经病理体征改变,免疫组织化学法检测血肿周围区HIF1-α的表达,以及参麦注射液治疗后的影响。结果:与模型组相比较,参麦组大鼠神经病理体征减轻明显(P<0.01);正常组和假手术组脑组织神经细胞无HIF1-α表达,而模型组血肿周围区HIF1-α的表达1天时增多,3天时达到高峰,5天时表达下降,至7天时仍有少量表达,与模型组相比较,参麦组各时间点的HIF1-α表达要少,二者有显著性差异(P<0.01)。结论:参麦注射液可能通过促进血肿吸收、抑制血肿周围区HIF1-α表达而发挥神经保护作用。Objective :To study The effect of Shenmai Injecton on the expression of hypoxia- inducible factor 1 -α( Hypoxia inducible factor 1 -α,HIF1 -α)in the perihematomal area in the Sprapue -Dewley Rat with intracerebral hemorrhage and its significance. Methods : The 50 rats were randomized into 4 groups : normal groups, false operation groups, modeling groups, and treating groups with Shenmai Injection,the rat both in the modeling groups and treating groups with Shenmai were made into four stages respectively in 1 d,3d,5d,Td after operation, the rat models were made by injecting Collagenase type Ⅲ stereotaxically;we observed the change of he pathologic sign of the nerve system and the expression of hypoxia - inducible factor 1 -α in the perihematomal area by immunohistochem,we also observed the effect of Shenmai Injecton on them. Results: Compared with modeling groups ,the pathologic signs of the nerve system were alleviated, hematoma were absorbed well ,the structure of neurons were relatively integrated( P 〈 0.01 ) ;there is no expression of HIF1 -α in the rats neurans of normal groups and false operation groups. However,the expressions of HIF1 -α in the rats neurans of the perihematomal area increased at lth day,get to a peak at the 3th day, followly decreased gradually at the 5th day, kept higher at the 7th day, there are obvious significant difference betwen treating group with Shenmai Injection and modeling groups( P 〈0.01 ). Conclusions: Shenmai injection may give a protective effect on the rats after ICH,which may be achived by promotting the absorption of hematoma and inhibting of the expression of HIF1 -α.
