摘  要:对不同取材时期、消毒处理、基本培养基和激素组合配比等因素对油桃"华光"、"曙光"和"秦光"茎尖离体初代培养和试管增殖倍数的影响进行了研究,结果表明,3种油桃茎尖培养最适宜的取材时期为新梢萌发期,萌发新梢的顶端嫩芽的污染率低,利于成活。用75%酒精30 s+0.1%升汞1 min+饱和次氯酸钠5min对油桃茎尖进行消毒处理效果最好。适合的初代培养基组合为:华光和秦光G+6-BA 0.50 mg/L+IBA0.50 mg/L,成活率分别为98.15%和72.34%;曙光G+6-BA 0.50 mg/L+IBA 0.125 mg/L,成活率为91.72%。适合的增殖培养基组合为:华光和曙光G+6-BA 2.00 mg/L+IBA 0.25 mg/L,平均有效芽数为2.36,2.89;秦光G+6-BA 1.50 mg/L+IBA 0.50 mg/L,平均有效芽数为3.16。Factors of different time and condition of the shoot tip, sterilization treatment, base medium and type of phytohormone and its combination which affected the shoot tip primary culturing and multiplying were studied. The results showed that in the shoot tip culture, the 75% alcohol 30 s and the 0.1% HgC12 1 min and the NaC103 5 min was the best for the nectarine shoot tip sterilization and the survival ratio was more high. The time of new shoot sprout was the best for gathering shoot tip, and the pollute rate was lower and adapt to develop. The G+6-BA0.50 rag/L+ IBA 0.50 mg/L was the best for the first culture for the Huaguang and the Qinguang, the survival rate was 98. 15% and 72.34%. The G+6-BA 0.50 mg/L+IBA 0. 125 mg/L was the best for Shuguang, the survival rate was 91.72%. The G+6-BA 2.00 mg/L +IBA0.25 mg/L was the best for the shoot tip multiply for the Huaguang and Shuguang, the No. of effective shoot was 2.36,2.89. The G+6-BA 1.50 mg/L + IBA0.50 mg/L was the best for the shoot tip multiply for the Qinguang, the No. of effective shoot was 3.16.
