卡马西平对大鼠海马脑片诱发场电位抑制作用的可能机制

摘  要:抑制剂量的卡马西平(CBZ)可抑制大鼠海马脑片的场电位使其幅值显著减小。非抑制剂量的CBZ(小于等于2×10-4mol/L)与γ-氨基丁酸(GABA.10-4,2×10-4,5×10-4和10-3mol/L)合用,抑制场电位的作用比单用GABA显著增强。用β-疏基丙酸10-4mol/L预处理脑片40-60min降低内源性GABA产量后,场电位幅值稳定地增大40%左右.此后再给抑制剂量的CBZ(3×10-4mol/L)对场电位无明显抑制作用,而CBZ与GABA合用则又显示了显著的抑制作用.上述结果表明,CBZ抑制场电位的作用可能是通过内源性GABA实现的。Carbamazepine (CBZ) 10-4and 2×10-4mol/L did not inhibitively affect the field potentials recorded from CA1 pyrarnidal cells,while CBZ 3 × 10-4 and 5-10-4mol/L distinctly depressed the amplitt1de of the field potentials. However, CBZ 2×10-4 mol/L together with GABA (10-4, 2 × 10-4, 5 ×10-4and 10-3 mol/L )brought about significantly stronger inhibition on the field potentials than that of GABA (same concentrations) alone. β-mercaptopropionic acid (MPA) 10-4 mol/L was used to preincubate hippocampus slices 40-60min,which could decrease the production of endogenous GABA and increase the amplittlde of field potentials by about 40%. If CBZ 3 × 10-4 mol/L was given after MPA, no distinct inhibition was observed; while this CBZ together with GABA (3 ×10-4mol/L) brought about strong inhibition. CBZ 5 ×10-4mol/L resulted in a similar outcome. The present results indicated that non-inhibitory dose of CBZ could facilitate the inhibitory effects of GABA on field potentials recorded from hippocampus slices, and inhibitory dose of CBZ might achieve its inhibition through endogenous GABA.
