摘 要:目的:探讨血管紧张素Ⅱ受体-1(AT1受体)在清醒WKY大鼠和自发性高血压大鼠(SHR)血压波动性(BPV)调节中的作用。方法:应用计算机化清醒自由活动大鼠血流动力学监测技术,分别观察了:(1)静注新型非肽类AT1受体拮抗剂Losartan(Los);(2)静注神经节阻断剂氯异吲哚铵(Chlor);(3)静注Chlor和Los;(4)静滴血管紧张素Ⅱ(ANGⅡ)及静注Chlor后静滴ANGⅡ对WKY大鼠和SHR血压和BPV的影响。结果:Los不影响WKY大鼠的血压,但减小其BPV,降低SHR的血压,但不影响其BPV。Chlor降低WKY大鼠和SHR的血压,增大它们的BPV。Chlor+Los可降低WKY大鼠和SHR的血压,但不影响它们的BPV。静滴ANGⅡ升高WKY大鼠和SHR的血压,增大它们的BPV;静注Chlor后再静滴ANGⅡ,两者血压均维持在各自基础值水平,而BPV都极显著增大。结论:交感神经调节的介入掩盖了Los对SHRBPV的影响;交感神经被阻断后,AT1受体的激活不仅对血压的维持有代偿作用,而且与此时血压发生剧烈波动,即BPV增大有关。Objective: To investigate the role of AT 1 receptor in the regulation of blood pressure variability (BPV)in conscious WKY rats and spontaneously hypertensive rats(SHR). Methods: Blood pressure(BP)was measured continuously by computerized system and analyzed for BPV in conscious freely moving rats: (1)after iv Losartan (Los); (2)after iv Chlorisondamine (Chlor); (3) after iv Chlor plus Los; (4) during intravenous infusion of ANGⅡ, and following Chlor during an infusion of ANGⅡ. Results: Los decreased BPV in WKY rats but not in SHR, and decreased BP in SHR but not in WKY rats. Chlor increased BPV and decreased BP in both strains. Chlor plus Los decreased BP, but did not influence BPV. ANGⅡ infusion increased BP and BPV. Following Chlor a continuous infusion of ANGⅡ maintained BP on the basal level whereas greatly increased BPV. Conclusion: The effect of Los on BPV is masked by sympathetic tone in SHR. Activation of AT 1 receptors are not only responsible for maintenance of BP but also for greatly increased BPV in both strains.
