HBV感染者外周血TLR4mRNA表达与疾病进展关系研究

摘　　要：构建pUCm-TLR4质粒作为定量模板,采用实时荧光定量RT-PCR的方法检测140例HBV感染者外周血单个核细胞(PBMC)中TLR4mRNA的含量,并结合HBV感染者临床情况进行分析。HBV感染者PBMC中TLR4mRNA的表达范围是1.1×105～10.5×107copy/μgRNA,显著高于健康对照组(4.5×105～1.25×106copy/μgRNA),其中慢性乙肝组TLR4mRNA的表达明显高于慢性重型乙肝组(P<0.05);HBV病毒载量的对数值与TLR4mRNA的含量存在负相关(r=-0.316,P<0.01)。结果表明,HBV感染者PBMC中TLR4mRNA的表达与疾病进展明显相关。pUCm-TLR4 plasmid was constructed as the quantitative template. By using real time fluorescence quantitative RT-PCR, the TLR4 mRNA levels in peripheral blood mononuclear ceils （PBMC） of 140 HBV infected patients were measured, The correlation between the TLR4 mRNA levels and clinical data of the patients was analyzed. The TLR4 mRNA levels in PBMCs of the HBV infected patients were from 1.1 × 10^5 to 10.5 × 10^7 copy/μg RNA, which obviously higher than healthy individuals （form 4.5 × 10^5 to 1.25 × 10^6 copy/μg RNA）. Furthermore, the TLR4 mRNA levels in PBMCs of the chronic hepatitis B patients were remarkably higher than that of severe chronic hepatitis B patients （ P 〈 0.05 ）. And the log value of HBV load in all the patients has negative correlation with the TLR4 mRNA levels in their PBMCs （r = - 0. 316, P 〈 0.01 ）. The results indicated that TLR4 mRNA level in PBMC was closely relative to the progression of hepatitis B.
