摘 要:目的构建一种新型纳米金界面压电石英晶体DNA传感器,探讨纳米金界面上进行寡核苷酸探针固定的最佳条件。方法构建纳米金颗粒修饰的石英晶振生物反应界面,进行巯基修饰的寡核苷酸探针的固定,对纳米金自组装pH值条件、探针固定浓度等影响因素进行实验研究;将构建的传感器和未经纳米金颗粒修饰的传感器用于铜绿假单胞菌检测,比较两种传感器的响应性能,并进一步对该纳米金界面压电石英晶体DNA传感器的再生进行研究。结果纳米金自组装的最佳pH值为6.0,探针最佳固定浓度为3.0μmol/L。纳米金颗粒修饰的石英晶振的响应性能大于未经修饰的石英晶振,可以重复使用5次。结论纳米金界面压电石英晶体DNA传感器,具有不需标记、操作简单、能实时在线检测、频率响应性能高、无污染和能重复使用等优点,在临床实验诊断方面应用前景广阔。OBJECTIVE To construct a novel piezoelectric quartz crystal microbalance (QCM) DNA sensor based on gold nanoparticle. METHODS The nanoparticles were immobilized on the gold surface of DNA sensor with selfassembly method. The pH value and concentrations of probes were optimized respectively. In addition, we detected the PCR products of Pseudomonas aeruginosa with both nanoparticle-based and common piezoelectric QCM DNA sensor array to assess the difference of frequency shift between the 2 sensors. RESULTS The optimum pH value for gold nanoparticle self-assembled was 6.0. The optimum immobilization yield was obtained at 3.0 μmol/L. The sensor could be reused for five times. CONCLUSIONS The nanoparticle-based QCM system we designed has the advantages of being rapid, label-free, and highly response performance and thus can be a useful supplement to clinical assay methods.
