摘  要:目的:构建转铁蛋白受体介导的Survivin反义RNA基因转移系统并观察其对肝癌细胞的体外杀伤效应及凋亡检测。方法:用EDC法制备抗转铁蛋白受体与多聚左旋赖氨酸(PLL)的复合物并用分子筛层析纯化。根据DNA阻滞实验结果Survivin反义RNA重组质粒与Ab-PLL按1:3混合使二者结合形成Ab-pLL-Survivin反义RNA(质粒DNA)复合物,将此复合物转染人肝癌细胞株HepG2细胞观察其对HepG2细胞的杀伤效应及细胞凋亡的影响。结果:转染复合物组HepG2细胞的增殖受抑制、凋亡明显增多。结论:本基因体系具有很好的靶向性及杀伤、促凋亡效应。Objective: To establish the surviving by transferring receptor, and to observe its special lethal effect apoptosis - induced effect on liver cancer cell. Methods : The conjugate of anti - TfR McAb and poly - L - lysine(PLL) was made by EDC and purified by molecular screen chromatography. According to the results of DNA block experiment, antisense surviving cDNA recombinant plasmid and McAb - PLL were combined by the ratio of 1:3, then the antisense survivin recombinant/McAb- PLL complex was used to transfer human hepatoma cell line HepG2, The lethal and apoptosis effects on HepG2 cell were observed. Results: HepG2 cellular proliferation was suppressed and its apoptosis increased in group with transduced complex. Conclusion: The gene system has better effect on targetly killing and improving apoptosis.
