摘 要:根据致病性嗜水气单胞菌的标志性毒力基因-气溶素(aerolysin)基因序列,设计出一对特异性引物,采用PCR方法,对22尾临床疑似患嗜水气单胞菌败血症的黄鳝进行了诊断,检测出其中17尾呈致病性嗜水气单胞菌阳性,检测过程仅需6h;而常规细菌分离鉴定则耗时72h,检出率仅为9/22。22 Monopterus albus with suspected Aeromonas hydrophila disease were diagnosised by PCR. The results showed that the detection rate of suspicious clinical samples was 17/22. The PCR assay that was established based on Aerolysin gene that was sign virulent gene of Aeromonas hydrophila 'which could detect the pathogenic Aeromonas hydrophila without biological methods of tox or serological method and could complete detection of samples in 6 hours. The same samples were diagnosised by isolation and verification of bacteria according to national standards, the deteetion rate of suspicious clinical samples was only 9/22,the detected procedure was complete with 72 hours. Compared with the method of isolation and verification, PCR assay is more sensitive,fast and precise, which dindicated that the method is suitable for investigation of epidemiology and detection of clinic sample on early-time infection, preventive and therapeutic measure ments can be taken in time, the assay has high practical value.
