摘 要:目的研究北美刺人参(Oplopanaxhorridus,OH)的HPLC指纹图谱,为科学评价及有效控制药材质量提供可靠方法。方法以紫丁香苷为对照品建立了O.horridus茎皮的HPLC指纹图谱,色谱条件为:色谱柱Kromasil C18column(250mm×4.6 mm,5μm),预柱Zorbax(12.5 mm×4.6 mm,5μm)流动相为乙腈-0.05%磷酸水溶液梯度洗脱,流速为1.0ml/min,柱温为35℃,检测波长210 nm。对11批茎皮药材进行了相似度分析,并同东北刺人参(O.elatusNakai,OE)茎皮指纹图谱进行了比较,采用系统聚类分析方法处理数据。结果相同种类药材的相似度较高,大于0.9,采用聚类分析可以聚为一类。结论指纹图谱可用于刺人参药材不同种类的鉴别。Objective To study on the HPLC fingerprints of Oplopanax horridus for offering a method on the quality control and assessment. Methods Syringenin was used as reference substance to establish' HPLC fingerprints of stem bark of O. horridus. Separations and determinations were carried out by linear gradient elution under following conditions: Kromasil C18 column(250 mm × 4.6 mm,5 μm), pre - column Zorbax ( 12.5 mm × 4.6 mm,5μm), acetonitrile - 0.05 % phosphoric acid solution as mobile phase, detection wavelength of UV at 210nm, flow - rate of 1.0 ml/min, column temperature of 35℃. Similarity calculation software was used to evaluate the whole HPLC spectrums of 11 samples ofrOH stem bark. Furthermore, it was compared with fingerprint of O. elatus Nakai stem bark and clustering analysis was applied for data analysis. Results The similarity of HPLC fingerprint was more than 0.9 among the same types of materials. Conclusion HPLC fingerprint can be used for differentiating sources of samples.
