摘  要:目的探讨血管内皮生长因子(VEGF)及环氧合酶-2(COX-2)在肝癌大鼠中的表达以及柔肝散结法对其表达的影响。方法40只雄性SD大鼠,随机分为空白组、模型组、青消方组、青消方防治组(简称"防治组"),每组10只。除空白组外,其余3组大鼠均以含0.05%2-AAF的饲料喂养12周,防治组在诱癌的同时灌服青消方。青消方组于诱癌第4周开始给药,剂量均为25 g.kg-1.d-1。12周末观察瘤体积及瘤重量的变化,并采用免疫组织化学方法检测各组大鼠标本中VEGF及COX-2的表达。结果模型组VEGF及COX-2的表达明显高于空白组(P<0.01);青消方组、防治组VEGF及COX-2的表达明显低于模型组(P<0.01);防治组优于青消方组。结论VEGF及COX-2在肝癌大鼠中表达增强,柔肝散结中药青消方能降低肝癌大鼠VEGF及COX-2的表达,早期治疗效果更佳。Objective To approach the expression of the VEGF and COX - 2 in rats with liver cancer and the effect of RouGan-SanJie Fa(RGSJF) on it. Methods 40 SD rats were divided into four groups at random:blank group, model group, QingXiao Fang group, FangZhi group, 10 rats in every group. Except for blank group, the other three groups were fed with bait vessel with 0.05% 2 - AAF for 12 weeks. FanZhi group was lavaged with QingXiao Fang when cancer was induced. QingXiao Fang group 25 g · kg · d^-1 was treated at the 4th week after cancer was induced. The volume and the weight of neoplasm were observed at the 12th week, and the expression of VEGF and COX -2 in every group was detected by immunehistochemical method. Results The expression of VEGF and COX-2 in model group was obviously higher than that of blank group (P 〈0.01 ). The expression of VEGF and COX -2 in QingXiao Fang group and FangZhi group was obviously lower than that of model group (P 〈 0.01 ), FangZhi group surpassed QingXiao Fang group. Conclusion The expression of VEGF and COX -2 was augmented in rats with liver cancer, QingXiao Fang can depress the expression of VEGF and COX -2 in rats with liver cancer, especially at earlier period.
