白英提取液对人肺癌A549细胞凋亡及Fas／FasL基因表达的影响

摘　　要：目的探讨白英提取液(STE)对人肺癌A549细胞凋亡及fas/fasL基因表达的影响。方法体外培养肺癌A549细胞,以0(正常对照组),10,20和40 mg/ml STE处理A549细胞48 h,并以25 mg/L顺铂(DDP)作为阳性对照。采用MTT法检测细胞增殖抑制率,TUNEL法检测凋亡率,半定量RT-PCR检测fas和fasL mRNA表达。结果与正常对照组比较,STE各浓度组细胞增殖抑制率显著上升(P(0.01),细胞凋亡率明显升高(P(0.01),基因fas mRNA表达显著上升(P<0.05或P<0.01),而fasL mRNA表达明显下降(P<0.05或P<0.01)。结论STE通过上调fas基因和下调fasL基因表达,诱导细胞凋亡,从而抑制A549细胞增殖。Objective To explore the effects of Solarium lyratum Thunb. Extract （STE） on induction of apoptosis and the expression of fas and fasL genes in A549 cells. Methods Lung cancer A549 ceils were treated with 0 （control group） , 10,20 and 40 mg/L for 48h, and the ceils were treated with 25 mg/L DDP as positive control. The proliferation inhibitory rate was evaluated by MTT assay. Induction of cell apoptosis rate was determined by TUNEL method. The expression of fas and fasL mRNA was detected by semi - quantitive RT - PCR. Results Compared with control group, the inhibitory rate was increased obviously （ P 〈 0.01 ）, the apoptotic rate was increased markedly （P 〈 0.01 ）. The expression of fas mRNA was increased significantly（ P 〈 0.05 or P 〈 0.01 ）, and fasL mRNA was decreased markedly （ P 〈 0.05 or P 〈 0.01 ） in STE groups. Conclusion STE can induce apoptosis by up - regulating expression of fas and fasL genes, and inhibiting the development of A549 cells.
