摘 要:目的探讨龙眼参黄酮(Longyanshen flavonoids,LF)对大鼠心肌缺血再灌注(myocardial ischemia reperfusion,MIR)引起的脂质过氧化损伤的影响。方法60只健康SD大鼠,随机分为假手术(Sham)组、缺血再灌注(MIR)组、生理盐水(nor-mal saline,NS)组和3个预处理组(LF低剂量组、LF中剂量组、LF高剂量组),各组大鼠雌、雄各半。结扎冠状动脉左前降支30min后,再灌注60min,建立MIR模型。用不同剂量LF在结扎冠状动脉左前降支前30min分别做预处理。再灌注结束后采血,测定血浆中谷草转氨酶(AST)、肌酸激酶(CK)、肌酸激酶同工酶MB(CK-MB)、超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)的改变,测量心肌梗塞范围。结果与MIR组比较,LF预处理能明显降低血浆中AST,CK,CK-MB和MDA含量,能提高SOD,GSH-Px活性,降低心肌梗塞范围。结论LF对大鼠MIR引起的脂质过氧化损伤具有显著的保护作用。Objective To investigate the influence of Longyanshen flavonoids (LF) on lipid peroxidation injury induced by myocardial ischemia reperfusion ( MIR ) in Spragne Dawley ( SD ) rats, Methods Sixty healthy SD rats were randomly divided into sham operation(Sham) group, MIR group, normal saline(NS) group,and three pretreated groups( LF low dose group, LF medium dose group and LF high dose group) ,with half female and half male rats in each group. MIR model was established by ligating left anterior descending coronary artery for 30 min followed by reperfusion for 60 min in rats. Different dosages of LF were administered 30 min before ligating left anterior descending coronary artery in every pretreated group, Blood was collected at the end of reperfusion for detecting the changes of aspartate aminotransferase (AST) , creatine kinase (CK) , creatine kinase isoenzyme MB ( CK - MB) , superoxide dismutase(SOD) , glutathione peroxydase ( GSH - Px) , malondialdehyde (MDA). The myocardial infarct size was measured after reperfusion. Results LF preconditioning could significantly decrease the contents of AST, CK, CK - MB and MDA, and the myocardial infarct size,and increase the activities of SOD and GSH - Px. Conclusion LF has a significant protective effect against lipid peroxidation injury induced by myocardial ischemia reperfusion in rats.
