体外培养的家兔视网膜色素上皮细胞凋亡模型的建立

摘　　要：【目的】探讨建立体外培养的家兔视网膜色素上皮(RPE)细胞凋亡模型的实验方法。【方法】实验组为加入浓度分别为0、100、200、400、600、800和1000μmol/L的吲哚美辛(I N),对照组为加入等体积无水乙醇,分别作用家兔RPE细胞24 h,用MTT法测量OD值来观察细胞凋亡的数量,对其进行定量分析。【结果】I N浓度分别为200、400、600、800、1000μmol/L作用RPE细胞24 h,其OD值分别为0.2094±0.0113、0.0799±0.0809、0.0685±0.0746、0.0538±0.0512和0.0501±0.0616,与对照组OD值0.3012±0.0395比较,都有显著性差异,有统计学意义。【结论】浓度≥200μmol/L的I N可以诱导RPE细胞有实验意义的凋亡。【Objective】To investigate the method of establishing the apoptosis model of cultured rabbit′s retinal pigment epithelium（RPE） cells in vitro.【Methods】Primary cultured and subcultured rabbit′s retinal pigment epithelium（RPE） cells were treated with indomethacin（IN） by concentration of 0,100,200,400,600,800 and 1 000 μmol/L for 24 h.Quantitative analysis of apoptotic rRPE cells was assessed by MTT method.【Results】The rRPE OD of≥200 μmol/L IN was 0.2094±0.0113,0.0799±0.0809,0.0685±0.0746,0.0538±0.0512 and 0.0501±0.0616 respectively and the OD of control sroup（0 μmol/L IN）was 0.3012±0.0395.【Conclusion】Indomethacin could induce apoptosis of cutured rRPE cells.
