摘 要:目的考察青龙衣多糖对S_(180)小鼠红细胞内Ca^(2+)和H^(+)的影响。方法建立肿瘤动物模型,分高、中、低剂量腹腔给予青龙衣多糖7d,采集红细胞,制备红细胞悬液。应用激光扫描共聚焦显微镜结合荧光探针Fluo-3/AM染色观察和测定S_(180)小鼠红细胞内Ca^(2+)的变化,结合荧光探针BCECF-AM染色观察和测定S_(180)小鼠红细胞内H+的变化。结果青龙衣多糖能降低S_(180)小鼠红细胞内Ca^(2+),与阴性对照组相比较有显著性差异(P<0.05);青龙衣多糖各剂量组均能显著降低S_(180)小鼠红细胞膜内的H^+浓度,与阴性对照组相比较有显著性差异(P<0.01)。结论青龙衣多糖组能维持S_(180)小鼠红细胞内阳离子浓度稳定平衡。Objective To study the effect of qinglongyi polysaccharide on cation concentration in erythrocyte of S180 mice. Methods A tumor mouse model was developed, and the mice were treated with high, medium, and low doses of qinglongyi polysaccharide through abdominal injection for 7 d. Erythrocytes were collected and erythrocyte suspensions were prepared. Change of Ca^2+ concentration in the erythrocytes ofS180 mice was measured by stained with Fluo-3/AM and observed with laser scanning confocal microscope, Change of H+ concentrations measured by stained with BCECF-AM and observed with laser scanning confocal microscope. Results Qinglongyi polysaccharide lowered the concentration of Ca2+ and H+ in erythrocytes of S180 mice, which is remarkable compared with negative control group(P〈0.05). Conclusion Qinglongyi polysaccharide can maintain the stability and balance of cation concentration in erythrocyte of S180 mice.
