摘  要:目的:建立宫内巨细胞病毒(CMV)感染的豚鼠模型,研究CMV宫内感染致异常妊娠结局的发生机制。方法:随机选择无CMV感染史的孕早期豚鼠,腹腔接种GPCMV,分娩24h内处死,运用套式聚合酶链反应(N-PCR)和地高辛标记的CMV晚期mRNA三相探针原位杂交技术,探究GPCMV宫内感染及其致异常妊娠结局的发生机制。结果:感染的孕鼠出现病毒血症征象,其子代出现流产、死胎等异常结局。N-PCR检测孕鼠及其子代GPCMV感染率分别为100.00%和94.57%,且感染脏器均出现非特异性组织病理学变化。杂交阳性信号位于子宫内膜基质细胞、子宫腺细胞、血管内皮细胞的胞质内。结论:孕早期GPCMV感染对妊娠结局影响最严重。中性粒细胞可将CMV携带到子宫毛细血管内皮细胞,导致胎盘感染和绒毛退行性变,这可能是进而引发子代生长发育异常的原因之一。Objective: To study the influence of the intrauterine GPCMV infection on the pregnant outcome by a guinea pig (GP) experimental model. Methods: GPs without infection were randomly selected, intraperitoneally inoculated with GPCMV during the first trimester. The pregnant GPs and their pups were killed within 24 hours after delivery. The infective status of the parental generation and filial generation and the outcome of pregnancy were observed by nest -polymerase chain reaction (N -PCR) and in situ hybridization (ISH) with three phases GPCMV late - mRNA probes labeled by digoxin. Results: The infective pregnant GPs had viremia manifestation. The abortion, fetal death and some other abnormal outcome were observed in their youngth. The infective rate of the parental generation and the filial generation were 100% and 94. 57%, respectively. All the visceras of the infective GPs appeared unspecific histological change. The positive signal of ISH was mainly distributed in the uterine endometrial stromal cells and microvascular endothelial cells. Conclusion: The GPCMV infection during the first trimester has a very important effect on the fetal growth and development. The intrauterine infection and the abnormal pregnant outcome are related with uterine infection.
