摘  要:采用荧光定量PCR方法,应用特异性引物,对淹涝胁迫下水稻叶片中低丰度表达的乙醇脱氢酶基因Adh2(Alcohol dehydrogene2)进行转录水平上的定量分析。实验得到一组基线平整、斜率大、指数区明显的S形荧光曲线。结果显示,在淹涝胁迫前期,Adh2基因表达持续增加,在水淹后8h表达量最大,随后下降。淹涝胁迫能诱导叶片中Adh2基因大量表达,且呈低-高-低的变化趋势。Real-time fluorescence quantitative polymerase chain reaction was used to determine transcription levels of Adh2 (Alcohol dehydrogene 2) gene in submerged leaves of rice using specific primers, expressed at low abundance under normal conditions. SYBR Green real-time PCR platform was established to check low abundance genes, which gave rise to a series of S curves with flat baselines, large slopes and distinct exponential areas. The results show that Adh2 transcription increases at the beginning of submergence and reaches the maximum level 8 hours after submergence, and then decreases. Adh2 transcription level in rice leaves follows a low-high-low pattern under submergence.
