摘  要:目的:采用蛋白质组学方法研究人骨髓间充质干细胞(hBMSCs)和成纤维细胞中的蛋白表达差异.方法:对hBMSCs和成纤维细胞的蛋白样本进行双向凝胶电泳分离,通过比较两种细胞的蛋白组学图谱,确定差异表达的蛋白点;而后对差异点进行基质辅助激光解析电离飞行时间质谱分析和蛋白数据库信息检索;最后选取其中的5种蛋白进行Western Blot实验验证蛋白质组学的研究结果.结果:在hBMSCs和成纤维细胞中鉴定出29种差异表达蛋白,其中有17种蛋白在两种细胞中有显著差别地表达,另外12种蛋白在两种细胞中有相同丰度地表达.Western Blot的实验结果进一步验证了蛋白质组学分析的结果.结论:hBMSCs与成纤维细胞中蛋白的差异表达体现了两种细胞在细胞形态、结构和功能上的异同性,这对于研究hBMSCs向成纤维细胞分化以及应用hBMSCs构建组织工程瓣膜具有重要意义.AIM: To find out the differences and similarities on the protein expressions between human bone marrow mesenchymal stem cells (hBMSCs) and human fibroblasts, which have different cell characteristics and similar cell morphology. METHODS: The protein extracts were obtained from hBMSCs and fibroblasts, and separated by two-dimensional gel electrophoresis (2-DE). And then the differential protein spots were identified by matrixassisted laser desorption ionization-time of flight-mass spectrometry (MALDI-TOF-MS). The results from simultaneous proteomic profiling were further validated by Western Blot of selected proteins. RESULTS: Twenty-nine differential proteins were successfully identified in hBMSCs and fibroblasts. Among them, 17 proteins were expressed significantly differently in hBMSCs from that in fibroblasts,and another 12 proteins equally expressed in the 2 types of cells. CONCLUSION: The differential protein expressions between hBMSCs and fibroblasts provide a new insight into the differential cell structures and functions between the 2 types of cells,which might be valuable for further study of fibroblasts dif- ferentiation from hBMSCs and the application of hBMSCs for creating tissue engineered heart valves in the future.
