摘 要:用非固定荧光标记的鬼笔环肽作为肌动蛋白探针观察并证明了丝瓜未萌发的花粉粒和不同生长时期花粉管中肌动蛋白纤丝的分布及其形态变化。又用细胞松弛素B(CB)、氯两嗪(CPZ)及N-乙酰马来酰胺(NEM)证明了丝瓜花粉管伸长与肌动蛋白既有密切的关系,也受Ca2+的调节。The localization and changes inform of actin cytoskeleton during pollengrain germination in Luffa cyindricawere visualized with FITC-phalloidinwithout fixation. The cytoplasm of theungerminated pollen grain conjoinedsome crystalline globular bodies (Fig.2), a storage form of actin. In hydrated pollen grain, numerous micro filaments (MFs) began to converge to-wards the germinant pore. After germination of pollen, the microfilament extended into the pollen tube. The globular bodies were transferred to the emerging pollen tube, gradually dissociated and then formed a network of axially oriented strands of microfilamentswith some branches. The microfilaments were distributed throughout thelength of the POllen tube, particularlyat the very tip where they formed adense region (Plate 11-8).Both the germination and growthof pollen tube were markedly inhibitedby cytochalasin B, N-ethyl maleimideand chloropromazine. The inhibitory effectS of the chemicals were probably theresult of regulation of Ca2+ on actinthrough calmodulin (Fig. 1 ).The effects of chemicals on thegrowth of POllen tube and the role ofactin in tip growth of POllen tube werediscussed.
