摘 要:目的:评估外用葡萄籽提取物原花青素对急性光损伤的Fas蛋白及Bcl-2蛋白表达的影响。方法:实验设计正常皮肤组、单纯2MED照射组、基质+2MED照射组、含葡萄籽提取物原花青素样品外用+2MED照射组共四组,连续3天同样处理24h后对皮肤取材,进行Fas蛋白及Bcl-2蛋白免疫组化染色。结果:含葡萄籽提取物原花青素样品外用+2MED照射组的Fas蛋白表达积分与单纯2MED照射比较明显下降,Bcl-2蛋白表达积分明显增加,接近正常皮肤。结论:葡萄籽提取物原花青素对日晒伤具有一定的防护作用,其机制可能是与Fas蛋白及Bcl-2蛋白表达相关。Objective In this study, the Influence of Grape Seed Proanthocyanidin Extract on Fas Protein and Bcl-2 protein expression from acute ultraviolet injury was investigated in skin by solar-simulator radiation. Methods Areas of skin of normal volunteers were divided into four sites: normal untreated skin, skin treated with 2-MED UV radiation, or skin treated with GPSE solutions or Vehicle 30minutes before 2-MED solar simulated radiation. Skin areas of different sites were examined immunohistochemically for Fas Protein and Bcl-2 protein expression. Results On immunohistochemical examination, Skin treated with GSPE before UV radiation reduced Fas protein expression and upregulated Bcl-2 protein expression compared with skin treated only with UV radiation. Conclusion Grape Seed Proanthocyanidin Extract may protect skin against acute sunburn partly through reducing Fas protein expression and upregulated Bcl-2 protein expression.
