肾上腺髓质素前体肽段(45-92)的心血管效应

摘　　要：目的：观察肾上腺髓质素前体肽段４５－９２的心血管效应。方法：离体大鼠血管灌流，离体心脏灌流和整体动物心血管功能测定。Objective: To investigate the effects of proAdm45-92-a putative product of adrenomedullin gene on cardiovascular system. Methods: The cardiovascular functions were measured in vivo and in vitro. Results:This study found that: (1) proAdm 45-92 significantly relaxed the rat isolated aortic ring precontracted by 5 ×10-7 mol/L norepinephrine in a dose-dependent and partly endothelium- dependent manner; (2 ) proAdm45 - 92(2. 5 x 10-8 mol/L) markedly increased coronary perfusive flow by (54. 4 ± 2. 3 ) % (P < 0. 05 ) and augmented the left ventricular systolic pressure(LVSP) by (21±4) % (P < 0. 05), but it had no influence on heart rate and LVdP/dtmax in the isolated perfused rat heart; (3 ) the mean arterial pressure was decreased [(1. 4 ±1. 0 ) kPa, P < 0. 05] by bolus injection of preAdm45-92 (5 nmol/kg) in anesthetized rats.Conclusion: ProAdm45-92 may be a vasodilative peptide; howevet, it seems to have no direct effects on myocardium in rats.
