摘 要:目的探讨增龄对大鼠膀胱、前列腺、后尿道α1A与α1D肾上腺素受体亚型分布的影响。方法2.5个月龄、8个月龄以及18个月龄3组大鼠各12只,取大鼠膀胱、前列腺、前列腺以远0.5cm后尿道。观察组织变化,并用RT-PCR、western印迹法测定各组织α1A和α1D受体mRNA水平及蛋白表达和分布变化。结果随着年龄增加,大鼠体重及膀胱、前列腺重量呈增加趋势,但各组间膀胱、前列腺重量无显著差异,而α1A与α1D肾上腺素受体在上述组织中的mRNA及蛋白水平的表达随大鼠年龄增加而显著降低。结论大鼠膀胱、前列腺、后尿道组织α1A与α1D肾上腺素受体表达与年龄呈负相关。临床上在使用α1受体阻滞剂治疗下尿路症状及前列腺增生患者时,可针对老年患者适当增加剂量。Objective To evaluate the influence of age on the expression of α1A and α1D adrenoceptor in lower urinary tract in rats. Methods A total amount of 36 rats at the age of 2.5 months, 8 months and 18 months (12 rats each) were enrolled in the study. The mRNA and protein level of α1 A and α1 D adrenergic receptor in the bladder,prostate and 0.5cm urethra distal to prostate was determined with RT-PCR and western blot relatively. Results The body weight increased significantly as the rat aging. There was no significant difference in the weight of bladders and prostates among the three groups. The α1A and α1D adrenergic receptor expressions in the bladder, prostate and posterior urethra were significantly downregulated in old rats. Conelusion The α1A and α1D adrenergie receptor levels in bladder,prostate and posterior urethra decrease as aging, According to this,the dose of α1 adrenoeeptor blocker should be increased in old patient in the clinical treatment of lower urinary obstruction and benign prostate hyperplasia.
