广谱根瘤菌NGR234在大翼豆根瘤中的分化和繁殖能力  

Differentiation and Reproductivity of Broad-host-range Rhizobium sp.NGR234 in Nodules of Macroptilium lathyoides

摘  要:利用盆栽法验证了广谱根瘤菌NGR234在大翼豆、白灰毛豆、扁豆、豇豆和大豆上的结瘤与固氮能力,用光学显微镜和电镜比较了其根瘤类菌体的形状,并采用稀释平板测数和显微微室培养法对大翼豆根瘤类菌体的繁殖能力进行了深入的比较研究。结果表明,根瘤菌NGR234不能在大豆上结瘤,在白灰毛豆上结无效瘤,能在扁豆、豇豆和大翼豆上结有效瘤,其根瘤类菌体以棒状类菌体为主。对大翼豆根瘤压碎汁的微室培养表明,不同瘤龄根瘤的类菌体的形态和繁殖率略有差异,杆状的根瘤小杆菌和多数棒状类菌体均能在人工培养条件下繁殖,其中类菌体的繁殖率随瘤龄的增加而增大,并在衰败前的根瘤中有明显的增加。The nodulation and nitrogen fixation ability of broad-host-range strain Rhizobium sp. NGR234 on Lablab purpureus ,Vigca unguculata,Macroptilium lathyoides, Tephrosia candida and Glycine max were confirmed by using plant pot experiment. Bacteroid differentiation was studied by using both of optical and electron microscopic techniques. Bacteroid reproductivity was compared by using viable count and microscopic incubation. It was demonstrated that NGR234 could form effective nodules on Lablab purpureus, Vigna unguculata and Macroptilium lathyoide, ineffective nodules on Tephrosia candida and fair to nodulate on Glycine max. Bacteroid of NGR234 was mainly in club shape in effective nodules. Results from viable count and microscopic incubation of crushed nodules in Macroptilium lathyoides demonstrated that shape and viability of bacteroids from nodules at different ages were different slightly. Both of small nods and majority of club shaped bacteroids eould reproduce in artificial incubation conditions. The reproduetivity of bacteroids was increased with the ages of nodules and significantly increased before nodule seneseing.
