摘 要:目的:探讨鹿茸多肽(PAP)对大鼠软骨细胞体外增殖的影响。方法:采用二次酶消化法获得大鼠关节软骨细胞进行传代培养并对第四代细胞进行药物干预,利用MTT比色、免疫组化检测增殖细胞核抗原(PCNA)、流式细胞术检测细胞周期等方法来检测各代次大鼠软骨细胞和药物干预后第四代软骨细胞的增殖情况。结果:大鼠软骨细胞随传代培养,MTT比色显示增殖呈下降趋势,免疫细胞化学检测PCNA表达减少,流式细胞术显示细胞增殖指数下降;PAP可以抑制这一趋势,促进传代培养的细胞增殖能力。结论:PAP对传代培养的大鼠软骨细胞的增殖能力有明显促进作用。Objective : The purpose of the present was to investigate the effect of pilose antler polypeptides (PAP) on the proliferation of rat chondrocyte in vitro. Methods : The rat articular chondrocytes were isolated by enzyme digestion and sub - cultivated in serial. The 4th chondrocyte was interfered with PAP. The chondrocytes of different generations and the 4th medicine intefered generation were detected with the methods of proliferating cell nuclear antigen , MTT( methyl thiazolyl tetrazolium) assay for proliferation and Flow cytometry for analyses of cell cycle distribution and PI ( proliferation index). Results : With ehondroeytes passage time increasing, the proliferating ability of chondrocytes descended. MTT assay showed that the proliferating ability of chondrocytes decreased, the expression of PCNA tapered in the experiment of immunohistochemistry, and PI descended in flow cytometry for analyses. PAP can inhibit the tendency and strengthen the proliferating ability of chondrocytes in serial subcultivation. Conclusion: PAP can significantly improve proliferation of chondrocytes in serial subcultivation.
