摘 要:目的初步研究禹白附的刺激性毒性成分。方法采用家兔眼结膜刺激性模型筛选禹白附刺激性成分,X-射线衍射法鉴别禹白附针晶,采用倒置显微镜、电镜扫描观测针晶结构性状,RP-HPLC法测定禹白附中草酸钙针晶含量,小鼠急性毒性实验验证针晶的刺激性毒性。结果各提取部位及草酸钙针晶的刺激性筛选表明禹白附的针晶具有强烈刺激性,该针晶经X-射线衍射鉴定为单斜晶系的CaC2O4.H2O晶体,测得禹白附生品的草酸钙针晶含量为0.584%,针晶具有显著的刺激性毒性,其浓度与家兔眼刺激程度呈量效关系。结论禹白附中所含的草酸钙针晶为其主要刺激性毒性成分。OBJECTIVE To investigate into the irritant toxicity of Rhizoma Typhonii.METHOD The irritant samples were produced using rabbit conjunctiva irritant model and -X-ray diffraction techniques was used to differentiate the needle crystals of Rhizoma Typhonii. With an inverted microscope, observation was made on the structure of the needle crystals; with RP-HPLC method, determination was carried out on the contents of calcium oxalate in Rhizoma Typhonii.RESULT The result showed that the needle crystals had strong irritability and was identified as oblique crystal CaC2O4·H2O. The content of calcium oxalate was 0.584%. There was a dose-effect relationship between the concentration of the needle crystals and the rabbit eye irritation. CONCLUSION The Rhizoma Typhonii needle crystal in Rhizoma Typhonii is the main irritant toxic component.
