摘  要:目的探讨血小板衍生生长因子(PDGF)及其受体(PDGFR)在HepG2细胞中的表达及其对细胞增殖及凋亡的作用。方法免疫组织化学法检测HepG2细胞PDGF及其受体的表达并予分型。经外源性PDGF作用后,用MTT法检测细胞增殖活性;流式细胞仪检测细胞凋亡和细胞周期分布;免疫组化半定量分析NF-κB和PDGFR表达情况,电镜观察细胞超微结构变化。结果HepG2细胞表达PDGF-A、PDGF-B及其受体PDGFR-α、PDGFR-β。PDGF-A及PDGF-B均可促进HepG2细胞增殖,抑制其凋亡,并促进NF-κB的表达,对PDGFR有上调作用,且PDGF-B作用强于PDGF-A。结论PDGF对肝癌细胞的发生发展起着重要促进作用。Objective To investigate the expression of PDGF and PDGFR in HepG2 cells and explore the role in its proliferation and apoptosis. Methods The expression of PDGF-A, B and PDGFR-α, β in HepG2 cells was detected by using immunohistochemical methods. HepG2 cells were stimulated with PDGF-A, B. The proliferative activity of cells was measured by MTT. FCM was used to assay the apoptosis of cells and the distribution in cell cycle. The expression of NF-κB and PDGFR was detected immunohistochemically, and the semi-quantification of the protein expression was analyzed by pathological image analysis software. Transmission electron microscope was applied to observe the ultrastructural changes of cells. Results PDGF-A, PDGF-B, PDGFR-α and PDGFR-β were identified in HepG2 cells. Both PDGF-A and PDGF-B could stimulate HepG2 proliferation, inhibited apoptosis of cells, and moreover, they could induce the expression of NF-κB and PDGFR, but the effect of PDGF-B was stronger than that of PDGF-A. Conclusion PDGF may play an important facilitate role in the hepatocarcinogenesis and its development.
