摘  要:目的研究异氟醚麻醉后大鼠海马蛋白质组的变化。方法将10只8月龄SD大鼠随机均分为异氟醚组(I组)和对照组(C组)。I组大鼠以1·2%异氟醚吸入麻醉2h,C组吸氧对照。于麻醉处理或吸氧对照结束后24h取海马组织行双向凝胶电泳和质谱分析以鉴定差异蛋白点。结果双向凝胶电泳结果显示I和C组差异蛋白质点共有17(4/13)个,MALDI-TOF-MS分析后鉴定出12种蛋白质。结论异氟醚麻醉可以引起大鼠海马蛋白质组变化,这些蛋白质变化可能具有一定的神经元保护作用。Objective To investigate the alteration of hippocampus proteome after isoflurane anesthesia in rats. Methods Ten 8-month-old SD rats were randomly divided into control group(group C)and isofluroame group(group I). The rats in group C only inhaled mixed gas containing 40% oxygen for two hours. The rats in group I were inhaled 1.2% isoflurane in 40% oxygen for two hours. The hippocampus of the rat was cut down for two dimensional gel electrophoresis (2-DE) 24 h after anesthesia. The different protein spots were analyzed by means of mass chromatographic analysis. Results There were 17 (4/13)different protein spots between group I and group C, of which 12 protein spots of all spots were identified by MALDI-TOF-MS. Conclusion The study shows that isoflurane may cause the alteration of hippocampal proteome in rats,and this change may be related to the neuron protection of isoflurane preconditioning.
