摘  要:目的:从生物化学、组织学和分子生物学角度观察4~6Hz振动对大鼠胃黏膜氨基己糖含量、黏蛋白5AC mRNA表达及其组织形态学变化,探讨振动对大鼠胃黏膜黏液分泌及形态的影响。方法:实验于2007-03/05在南京农业大学农业部动物生理生化重点开放实验室进行。取健康成年SD大鼠30只,随机分成对照组和4,5及6Hz振动组,对照组6只,其余3个组各8只,分别给予0,4,5及6Hz振动2h后立即断颈处死,取胃黏膜及胃腺区组织,测定胃黏膜氨基己糖含量、检测黏蛋白5AC mRNA表达,观察其胃黏膜形态学变化.结果:30只大鼠均进入结果分析,无脱失。①大鼠胃黏膜氨基己糖含量:随着振动频率的增加而下降,4Hz振动组与对照组相比无显著变化,而5,6Hz振动组显著低于对照组(P<0.05,0.01)。②黏蛋白5AC mRNA表达:5Hz振动组高于对照组(P<0.01),而6Hz振动组与对照组相比无显著差异(P>0.05)。③胃黏膜形态:5,6Hz振动组大鼠胃黏膜上皮不完整,上皮及固有层细胞脱落。结论:5Hz和6Hz的振动会导致大鼠胃黏膜损伤,黏液分泌减少,并且在此过程中可能存在应激的代偿效应。AIM: From the views of biochemistry, histology and molecular biology, this study observed the effects of 4-6 Hz vibration on concentration of hexosamine, the expression of mucin (MUC) 5AC mRNA in gastric mucosa and histomorphology, and investigated the effects of body vibration on rats' gastric mucus secretion and mucosal structure. METHODS: Experiments were performed at the Key Open Laboratory of Physiology and Biochemistry of Ministry of Agriculture of Nanjing Agricultural University from March to May 2007. Thirty healthy adult SD rats were randomly divided into four groups including control (n=6), 4 Hz (n=8), 5 Hz (n=8) and 6 Hz (n=8) vibration groups. All the rats were killed after they were quivered continuously for 2 hours except for control ones. The gastric mucosa and gastric gland tissue of rats was immediately attained. The concentration of hexosamine and the expression of MUC5AC mRNA in gastric mucosa of each rat were tested. The tissues structure of gastric mucosa was investigated from those rats. RESULTS: Thirty rats were included in the final results, no drop out. (1)The concentrations of hexosamine from gastric mucosa of rats decreased with the increase in frequency of vibration. There were no significant changes between 4 Hz vibration group and control group. However, it was lower in the 5 Hz and 6 Hz vibration groups than in the control group (P 〈 0.05, 0.01). (2)The expression levels of MUC5AC mRNA of rats gastric mucosa in 5 Hz vibration group were higher than in the control group (P 〈 0.01). but insignificant variance was shown between 6 Hz vibration group and control group (P〉0.05). (3)Incomplete mucosal epithelium and desquamation were observed in rats of 5 Hz and 6 Hz vibration groups. CONCLUSION: Gastric mucosa will be injured and the mount of mucus secretion will induce after the rats were vibrated by 5 Hz and 6 Hz whole body vibration. Stress compensation effects appear at the same time.
